Synthesis of novel chiral sulfoxides, its purification, characterization and study of enantioseparation in high-performance liquid chromatography.

The major goal of the master thesis was the synthesis of a novel chiral sulfoxide, its purification, spectroscopic characterization and study of enantioseparation using polysaccharide-based chiral stationary phases in high-performance liquid chromatography. In the frame of the current thesis Promethazine Hydrochloride was oxidized to Promethazine sulfoxide via three different ways: A) using 3-chlorperbenzoic acid as an oxidizing agent at 00 C, in a homogeneous system. Chloroform was used as a solvent. B) using 3-chlroperbenzoic acid as an oxidizing agent at 00 C in a heterogeneous system in water as a solvent. C) using hydrogen peroxide at 00 C in a homogeneous system in water as a solvent. The final product was identified and characterized using UV-VIS, IR and NMR spectroscopy. Separation of enantiomers of the obtained substance was performed in high-performance liquid chromatography using cellulose and amylose derivatives as a chiral selectors.
